Gleeman Vox
Otto Destruct
Percival Tachyon
Captain Slag
Angstrom Darkwater
Cassiopeia
These are just villains that either are still alive or you didn't see die, I didn't include Dr. Nefarious as he was isn't that far from the past but if you ask me I would say Drek.
But as the humor of Insomniac games they would probably have someone like the Qwark Fan boy which would be funny.

Of those on your list, it can't be either Drek or Tachyon, as IG confirmed on their forums it's no main villain of the past.
And I doubt it's from High Impact Games' games, as those aren't that well-known and it's IG making the game after all.
Plus, Drek, Vox, Darkwater and Cassiopeia are dead.
Maybe it's a minor villain, or even an ally that turned bad.

Nope, Ratchet's on Clank's back so it is Giant Clank.

The villain is interested more on Qwark than on Ratchet so I think Slag wouldn't do it as he wants to kill Ratchet. I think he doesn't even know who Qwark is.
